Stiamo celebrando la festa della Santa Croce. Ritrovata dall’imperatore Constantino che indisse percio’ la festa della Santa Croce. Festa che fu celebrata per la prima volta nel luogo della crocifissione e nel luogo della risurrezione di Gesu Cristo.
Sant’Elena fece portare una malata a contatto con la croce, che guari’ miracolosamente. Da quel giorno in ricordo il 14 settembre e’ il giorno della Festa della Santa Croce.
La Croce e’ il prezzo del nostro riscatto pagato da Gesu Cristo stesso a costo della sua vita. La luce di Cristo e’ quella che illumina il nostro cammino. La difficolta’ e’ capire che le nostre croci possono diventare per noi le chiavi del paradiso, dell’immortalita’ e della salvezza.
La mentalita’ di oggi e’ quella di sbarazzarci delle croci. Ma questa via non ci salva e non puo’ garantirci il paradiso!
Gesu’ Cristo e la sua Santa Croce sono quindi la nostra salvezza!
Sia lodato Gesu’ Cristo!

We are celebrating the feast of the Holy Cross.
Found by the Emperor Constantine who therefore announced the feast of the Holy Cross. Feast that was celebrated for the first time in the place of the crucifixion and in the place of the resurrection of Jesus Christ.
Saint Helena had a sick woman brought into contact with the cross, which miraculously healed. From that day in remembrance, September 14 is the day of the Feast of the Holy Cross.
The Cross is the price of our ransom paid by Jesus Christ himself at the cost of his life.
The light of Christ is the one that illuminates our path. The difficulty is understanding that our crosses can become for us the keys to heaven, immortality and salvation.
Today’s mentality is to get rid of the crosses.
But this route does not save us and cannot guarantee us paradise!
Jesus Christ and his Holy Cross are therefore our salvation!

Iniziamo una settimana spèciale. Questa S. Messa e’ dedicata alla SS. Trinita’.
L’amore, la misericordia sono caratteristiche del Padre. Il Perdono, la Grazia del Figlio. La Comunione e’ invece dello Spirito Santo.
L’unita’ e’ attorno alla verita, che pertanto e’ soltato quella di Cristo (“Io sono la Verita’ e la Vita”).
Solo Dio puo’ riportare la Chiesa all’unita’ e alla purezza dei primi secoli. Negli ultimi decenni i tentativi ecumenici della Chiesa sono stati puramente umani. E hanno portato inevitabilmente al fallimento della falsa chiesa.
Dobbiamo pregare Dio! Il resto ci sara’ dato in piu’.
Sia lodato Gesu’ Cristo!

Let’s start a special week. This Holy Mass is dedicated to the SS. Trinity’.
Love, mercy are characteristics of the Father. Forgiveness, the Grace of the Son. Communion is instead of the Holy Spirit. Unity is around the truth, which therefore is only that of Christ (“I am the Truth and the Life”).
Only God can restore the Church to the unity and purity of the first centuries.
In recent decades, the Church’s ecumenical endeavors have been purely human. And they inevitably led to the failure of the false church. We have to pray to God! The rest will be given to us in more.

Signore Gesù, io credo che sei presente realmente nel Santissimo Sacramento dell’altare, in corpo, sangue, anima e divinita’. Ti amo sopra ogni cosa e Ti desidero nell’anima mia. Poiché ora non posso riceverti sacramentalmente, vieni almeno spiritualmente nel mio cuore. Come già avvenuto, io Ti abbraccio e mi unisco totalmente a Te. Non permettere che io mi separi mai più da Te. Cosi’ sia.
Eterno Padre ti offriamo, tramite l’intercessione della Vergine Maria, il Corpo e il Sangue Preziosissimo di Gesù Cristo per il perdono dei nostri peccati, in suffragio dei nostri defunti, delle anime del Purgatorio e per i bisogni della Santa Chiesa.
Gesù, ti adoro in ogni luogo dove abiti nel Santissimo Sacramento; ti faccio compagnia per coloro che ti disprezzano, ti amo per coloro che non ti amano, ti do sollievo per coloro che ti offendono. Gesù, vieni nel mio cuore!

Oggi e’ la festa del santissimo nome di Maria.
Il vangelo di oggi e’ una pietra miliare: chi si loda sara’ umiliato, chi si umilia sara’ esaltato. Subito ci viene in mente il nome di Maria, lei che si e’ umilata e noi. Ma oggi il nome di Maria risuona sulla terra e nei cieli. Lei e’ potente contro i demoni! Lo sappiamo noi stessi quando l’abbiamo pregata con l’Ave Maria!
Ma il nome di Maria e’ anche oltraggiato! Molti di noi la offendono. Dobbiamo avere riguardo del suo nome e ringraziarla dei suoi doni. La nostra preghiera e’ sempre piena di preoccupazioni. Maria ci sta rimproverando come figli. Rivolgetevi a me con fiducia, non preoccupatevi per il futuro! Non possiamo sapere cosa ci riserba Dio per il futuro. Ma non dobbiamo pregare con ansieta’. Dio non ci sottopone mai a prove eccessive per nostre forze (prove che Lui sa non possiamo superare). No mai!
Nel Vangelo Gesu’ guarisce un idropico (malattia che porta a gonfiare il corpo). Vuole guarire non solo la malattia del malato ma anche la superbia dei farisei commensali e ospiti.
Il Figlio di Dio puo’ guarire anche il Sabato, che e’ appunto il giorno di Dio per gli ebrei! Lui e’ Dio e’ puo’ guarire anche il Sabato e la Domenica, il giorno del Signore Gesu’ Cristo.
Occorre quindi piu’ fede. Dobbiamo esporci alla sua grazia, invocando sempre il nome di Maria!

Today is the feast of the most holy name of Mary.
Today’s Gospel is a milestone: whoever praises himself will be humbled, whoever humbles himself will be exalted. The name of Mary immediately comes to mind, she who humbled herself and us.
But today the name of Mary resounds on earth and in the heavens. She is powerful against demons! We know it ourselves when we prayed to her with the Hail Mary! But the name of Mary is also outraged! Many of us offend her. We must be considerate of her name and thank her for her gifts. Our prayer is always full of worries. Maria is scolding us as children. Contact me with confidence, don’t worry about the future!
We cannot know what God has in store for the future. But we don’t have to pray anxiously. ‘ God never subjects us to excessive tests on our strength (tests that He knows we cannot overcome). Never!
In the Gospel, Jesus heals a dropsy (a disease that leads to swelling of the body). He wants to heal not only the sickness of the sick but also the pride of the Pharisees who are diners and guests.
The Son of God can also heal on Saturday, which is precisely God’s day for the Jews! He is God and can also heal on Saturday and Sunday, the day of the Lord Jesus Christ. Therefore, more faith is needed. We must expose ourselves to his grace, always invoking the name of Mary!

‘Non si vaccina in presenza di varianti, perché sarebbe come trattare una persona affetta da un’infezione batterica con un basso livello di antibiotici’
Quanto incide la vaccinazione sulla possibilità che il virus muti in varianti resistenti? La domanda, più volte posta in dibattiti tra favorevoli e contrari alla vaccinazione, ha una risposta complessa e articolata. Tutto dipende infatti dal livello di trasmissione del virus nel momento in cui si inizia a vaccinare.
A fare un quadro chiaro della realtà, era stato appena due mesi fa il microbilogo Andrea Crisanti, in una dichiarazione a Uno Mattina del 19 aprile 2021, poi riportata da diverse agenzie (qui).
‘La probabilità che una variante emerga è direttamente proporzionale al livello di trasmissione – aveva spiegato Crisanti citando le 92 varianti Covid presenti in quel momento in Brasile -. Se a questo dato sovrapponiamo la vaccinazione si crea la situazione in cui si selezionano le varianti resistenti al vaccino. Non si vaccina con alti livelli di trasmissione, questo è l’Abc della genetica. Più c’è moltiplicazione virale più crescono le probabilità che aumentino varianti e che possa emergere una variante resistente al vaccino. Non si vaccina in presenza di varianti, perché sarebbe come trattare una persona affetta da un’infezione batterica con un basso livello di antibiotici o addirittura con antibiotici sbagliati. Così si selezionano le varianti resistenti, è inevitabile’.
Una lezione chiara pronunciata da uno scienziato decisamente favorevole alla vaccinazione. Una lezione che però appare oggi, a soli due mesi distanza e in piena emergenza di variante Delta, dimenticata.

For the adequate design of COVID-19 vaccines, we must consider the selection of antigens, the proper selection of the platform for antigen production, adjuvants and other substances used during vaccine formulation. However, the success of the SARS-CoV-2 vaccines will also depend to a large extent on the production capacity to meet the volumes required by a global pandemic, an adequate distribution of doses, correct administration regimens, as well as appropriate use according to the type of vaccine, which could contribute to combat SARS-CoV-2 emerging variants.
Because the generation of new variants is aleatory, the longer the pandemic lasts, the higher the probability for the generation of new variants. Therefore, controlling rapidly the pandemic worldwide by a combination of a well-organized vaccination campaign coupled with strict sanitary measurements are key factors to prevent extending the pandemic for another year or even longer.
The S glycoprotein has been the main target for the development of vaccines against SARS-CoV-2 (4) and has been shown to be efficient enough to protect individuals who are vaccinated with this antigen or highly immunogenic regions of the same, such as the RBD (188). Companies such as Pfizzer & Biontech, AstraZeneca, Novavax, CanSino Biological, and Inovio Pharmaceuticals, have chosen to use the complete S glycoprotein, each following different strategies modifying the structural characteristics of the S glycoprotein, with the aim of improving its expression, recognition, stability or immunogenicity (67, 74, 78, 79, 150). Alternatively, other companies or academic institutions have chosen to use highly immunogenic regions within the S glycoprotein, such as the RBD. RBD-based vaccines may present modifications that improve their presentation, including the incorporation into VLPs (101), trimerization motifs (103), domain duplications (102) and even fusion carrier proteins (104). In addition, other types of vaccines such as those based on epitopes from the S glycoprotein are currently tested in animals models (112).
The wide variety of proposals approved and in clinical phases allows several options to design the best vaccination strategies against the SARS-CoV-2 (6). However, the emergence of new variants casts doubts on the effectiveness of the vaccines developed so far (174). It has been shown that some mutations present in new variants escape neutralization, particularly mutations found around the RBD (171–173). Given these mutations in the RBD, some vaccines have shown moderate neutralizing effects with variants such as B.1.351 and P.1 (150, 152, 158, 159). So far, only vaccines based on full-length S glycoprotein have been evaluated regarding their ability to neutralize new SARS-CoV-2 variants (182). It is imperative to evaluate the effectiveness of all vaccines against new variants if we intend to control the pandemic and prevent COVID-19 from becoming an endemic disease. Still, much like influenza, we cannot discard the possibility of having to generate new vaccines against emerging SARS-CoV-2 variants every year and implement frequent vaccination programs.
We consider that due to the emergence of new SARS-CoV-2 variants in countries with limited access to vaccines, the redesign SARS-CoV-2 vaccines based on these new variants is highly recommended.
We propose that the redesign of SARS-CoV-2 vaccines based on S glycoprotein, RBD or its epitopes should be considered for their proper use according to the geographic distribution of the SARS-CoV-2 variants. For example, epitope-based vaccines could have a lesser effect in countries with high prevalence of variants with immune escape, such as that observed with variants B.1.351 and P.1.
We should take full advantage of strategies such as genomic databases, structure prediction systems, and predictors of antigenic determinants in the design of vaccines against emerging variants (189), which may be useful assets to predict conformational and linear epitopes that can be recognized by B lymphocytes (190, 191). Most of the vaccines in use or in more advanced clinical trials utilize the complete SARS-CoV-2 virus, the full-length S glycoprotein, the RBD domain or RNA encoding the S glycoprotein (Tables 1 and 2).
Biomaterials are defined as substances of synthetic or natural origin, which can interact with living systems, in which they can perform therapeutic or diagnostic functions in order to improve the quality of life of individuals (192). The use of biomaterials in the development of vaccines against SARS-CoV-2 has been widely extended including: VLPs, DNA, RNA, liposomes, viral vectors, among others (Tables 1 and 2). However, the use of biomaterials in combination with peptides and predicted epitopes has not been sufficiently explored. There are biomaterials that form nano and microstructures which have been shown efficacy as antigen delivery systems, as enhancers of the immune response (adjuvant) and as antigen stabilizers (193, 194). Polymers and copolymers (195, 196), self-assembling proteins and self-assembling peptides (SAPNs and SAPs, respectively) (197, 198), microneedles (199), metallic nanoparticles (199, 200) and carbon nanomaterials (200). All of the previous mentioned biomaterials should be extensively explored in new generation of SARS-CoV-2 vaccines.
Conclusion
We must recognize that the design and development of efficient vaccines against SARS-CoV-2 is the best strategy to combat COVID-19, fortunately many of them have shown high efficiency. Nevertheless, the emergence of new variants can jeopardize the success achieved so far with vaccination. To reach total control of the COVID-19 pandemic, the combination of several strategies will be necessary, which include an adequate distribution of SARS-CoV-2 vaccines taking into consideration the geographical distribution of the variants, the re-designing or SARS-CoV-2 antigens as well as the use of other technological tools such as bioinformatics and the use of novel biomaterials.

Se i ricercatori avessero tentato di prevedere quale variante di coronavirus avrebbe conquistato il mondo, la variante Delta non sarebbe stata la loro prima ipotesi. Ma dalla sua prima apparizione in India nel dicembre 2020, questa variante altamente contagiosa è diventata il ceppo predominante del virus, responsabile di oltre il 90 per cento dei nuovi casi di COVID-19 negli Stati Uniti [l’Italia ha una percentuale simile, NdR].

In un recente studio di preprint, non ancora pubblicato su una rivista scientifica, l’epidemiologo Nathan Grubaugh della Yale School of Public Health di New Haven, in Connecticut, ha testato in che modo 18 diverse varianti reagivano al siero prelevato dal plasma – la componente liquida del sangue – di 40 operatori sanitari completamente vaccinati. Ha scoperto che gli anticorpi dei partecipanti erano in grado di neutralizzare bene la variante Alfa e abbastanza bene Delta. Ma erano meno efficaci contro le varianti che portano le mutazioni E484K, come Beta o Gamma, identificate per la prima volta in Sudafrica e Brasile, rispettivamente.
I risultati sono stati sorprendenti, considerato che Delta è stata molto più efficace nel diffondersi rispetto a Beta o Gamma. Mehul Suthar, virologo della Emory University di Atlanta, afferma che potrebbe esserci un limite al numero di mutazioni che un virus può acquisire sulla proteina spike prima che non sia più in grado di legarsi al recettore ACE2. “Ci sarà sempre questo tiro alla fune tra il virus che guadagna la capacità di contagiare, diffondersi e replicarsi e la sua capacità di sfuggire alla risposta anticorpale”, aggiunge.
Suthar sottolinea che la variante Kappa, emersa in India nello stesso periodo di Delta, condivide la maggior parte delle mutazioni di Delta e una mutazione simile a E484K. Ma Kappa non si è diffusa in tutto il mondo, suggerendo che queste mutazioni possano interagire in un qualche modo che rende il virus meno adatto evolutivamente. “È stata un po’ una sorpresa che sia stata la variante Delta a spiccare il volo”, spiega Suthar. “In realtà è stato confortante.”
Grubaugh pensa che sia improbabile che appaiano nuove mutazioni significative della proteina spike. Invece, il virus più adatto avrà una combinazione delle mutazioni “migliori” che permetteranno al patogeno di diffondersi ampiamente tra la maggior parte delle persone. Ma è difficile prevedere quali saranno. “Penso che abbiamo una conoscenza abbastanza buona di quali mutazioni dovremmo cercare, ma questo non significa necessariamente che se le vedessimo, sapremmo esattamente come si comporteranno”, dice.
Cooper è d’accordo. “È logico che quella combinazione di alta trasmissibilità e capacità di eludere gli anticorpi si evolverà, se possibile, a condizione che il numero di infezioni rimanga alto”, afferma. Più persone rimangono non vaccinate, più alta è la possibilità che emerga qualcosa di peggio di Delta.
Un virus potenzialmente da incubo è apparso l’anno scorso: una variante chiamata B.1.620 che è stata osservata per la prima volta in Africa. Portava la mutazione E484K, insieme a una serie di altre mutazioni della proteina spike che potrebbero aumentare la trasmissibilità. Con questa sola mutazione, la reazione sarebbe: “Oh mio Dio”, afferma Emma Hodcroft, ricercatrice in campo genetico all’Università di Berna, in Svizzera. Ma il numero di casi di B.1.260 è presto diminuito. “È chiaro che non è così semplice avere tutte queste mutazioni ed essere la cosa peggiore in giro”, sostiene Hodcroft.
La buona notizia è che sarebbe relativamente semplice modificare i vaccini a mRNA prodotti da Pfizer e Moderna per renderli più efficaci contro tale variante, sottolinea Nathaniel Landau, microbiologo del Langone Medical Center della New York University. “Se emergesse una variante in grado di sfuggire meglio di quella attuale, si avrebbe davvero bisogno di iniezioni di richiamo contro quella variante”, spiega.
Nel frattempo, però, i vaccini attuali rimangono il modo migliore per prevenire l’emergere di nuove varianti. “È chiaro che, con questi virus, non è che sconfiggendo la variante Delta sconfiggeremmo la pandemia”, dice Suthar. “Ce n’è un’altra che continua a emergere.” Se troppe poche persone si vaccinano e il virus si diffonde senza controllo – conclude – ci sarà solo questo ciclo infinito di infezione, mutazione e trasmissione”.
